{"created":"2023-06-19T06:41:44.552103+00:00","id":74,"links":{},"metadata":{"_buckets":{"deposit":"90b68338-1fbc-419f-a52c-5b6f2146dc70"},"_deposit":{"created_by":2,"id":"74","owners":[2],"pid":{"revision_id":0,"type":"depid","value":"74"},"status":"published"},"_oai":{"id":"oai:opcnr.repo.nii.ac.jp:00000074","sets":["1:10"]},"author_link":["231","15"],"item_10002_biblio_info_7":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2003-03","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"4","bibliographicPageEnd":"124","bibliographicPageStart":"118","bibliographic_titles":[{"bibliographic_title":"沖縄県立看護大学紀要"},{"bibliographic_title":"|isolated island@@@Yo-Kaigo Elders@@@The Long-Term Care Insurance System@@@In-home services@@@in-island completion service system","bibliographic_titleLang":"en"}]}]},"item_10002_description_5":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"昨年度の本誌に、scrape-loadingによって蛍光色素を細胞内に取り込ませ、周囲細胞への移行の様子で、gap-junctionを介する細胞間の情報交換について検討するEl-Foulyらの手法について紹介すると共に、この方法を用い、腎上皮由来の細胞と言われるNRK-52E細胞で得られた結果の幾つかについて報告したが、今回は、1つの細胞に、直接選択的に、蛍光色素を注入する事のできる微量注入装置を用いて、蛍光色素を直接細胞内に注入する方法で行った実験の結果について報告する。細胞は、昨年同様、NRK-52E細胞であり、細胞間情報交換の指標とした蛍光色素も同じくlucifer yellow(LY)である。色素が注入された細胞を明らかにする為、LYと共にethidium bromide(EB)を細胞内に注入した。微量注入装置の自動注入機能を利用し、保持圧を150 ヘクトパスカル、注入圧を800-1100ヘクトパスカル、注入時間を0.8秒に設定して注入を行った。その結果、NRK-52E細胞群では、1つの細胞に注入された色素は、ほぼ均等に周囲の細胞に拡がること、10-15分で、色素の拡がりはピークに達する事、代表的な腫瘍プロモーターであるTPA(12-O-tetradecanoylphorbol-13-acetate)(10-100 ng/ml)、10-20分処置によって、色素の拡がりが完全に抑制されることがわかった。\nIntercellular communication through gap junction was studied in NKR-52E cells in culture using the transfer of a fluorescent dye, lucifer yellow, as a measure. Instead of a scrape loading method used in our previous work(Une K. and Imai S.), a direct microinjection method was used. The dye was injected with a microinjector(Eppendorf FemtoJet) via a glass capillary (Eppendorf Femtotips) impaled into the cell. The microinjector was operated with a micromanipulator(Eppendorf 5171). Another dye, ethidium bromide, was injected together with LY to identify the cell into which the dye was injected. Fluorescence of the dyes was monitored with an inverted fluorescent microscope (Leitz DMIRB).
